Formalin is a commonly used fixative for tissue preservation in histopathology labs. Formalin or other aldehyde fixation forms protein cross-links that mask the antigenic sites in tissue specimens, thereby giving weak or false negative staining for immunohistochemical detection of certain proteins. The HIER solution is designed to break the protein cross-links, therefore unmask the antigens and epitopes in formalin/aldehyde-fixed tissue, thus enhancing staining intensity of antibodies. Our HIER is highly effective for free floating section, slide-mounted cryostat sections and paraffin-embedded slide-mounted sections. Our best results were obtained with a 30 min incubation in this preheated HIER and maintained at 80-100 °C, followed by 30 min incubation in 5% Normal Goat Serum or Normal Donkey Serum to reduce non-specific staining.
