This experiment introduces the use of restriction enzymes as a tool to digest DNA at specific nucleotide sequences. Bacteriophage lambda DNA has a linear structure and 6 Eco RI recognition sites. Separation by agarose gel electrophoresis of an Eco RI digest of lambda DNA will yield 6 bands (5 distinct bands, two are very close in size) corresponding to the DNA fragments. Kit includes: instructions and Ready-to-Load dye samples for 24 gels. All you need: electrophoresis apparatus, power supply, automatic micropipet and tips, balance, microwave or hot plate, 65 degree C waterbath, visualization system. EDVO-Kit 112-C contains pre-digested DNA samples ready for electrophoresis sufficient for twenty-four (24) agarose gels with the use of an automatic micropipet. A: Standard DNA Fragments, B: Lambda DNA cut with Eco RI, C: Lambda DNA (uncut), Practice Gel Loading Solution, 100ml plastic graduated cylinder (packaging). Experiment components include enough sample for 24 gels if you are using an automatic micropipet for sample delivery. Use of transfer pipets will yield fewer gels. Reagent quantities are based upon the use of Horizontal gel electrophoresis apparatus, Model M12 and sample volumes aliquoted at 40 �l each.
